Product Specific Information

The 93 monoclonal antibody recognizes an epitope shared by mouse CD16 and CD32.
CD16 (FcγRIII) and CD32 (FcγRII) are low-affinity IgG Fc receptors expressed by B cells, macrophages, NK cells, and neutrophils.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometric analysis of mouse splenocytes (≤1.0 µg/test in 100 µL volume).
Cell concentration: 10⁵–10⁸ cells/test. Optimize antibody titration for best performance.

Important Notes:

  • NovaFluor dyes are incompatible with DNA-intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
    Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
    Add 5 µL per sample to antibody cocktails prior to cell addition.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).
  • NovaFluor conjugates use Phiton™ technology for engineered fluorescence with minimized spillover and spread.
  • Excitation: 552 nm; Emission: 690 nm; Laser: 561 nm (Yellow).
  • Non-specific staining observed in SJL mice B cells; not observed in BALB/c or C57BL/6 mice.

Target Information

CD16: 50–65 kDa surface molecule; exists in transmembrane and PI-linked forms. Expressed by NK cells, some T cells, and granulocytes. Mediates ADCC via FcγRIII.

CD32: 40 kDa glycoprotein; low-affinity receptor for IgG (FcγRII).
Functions include endocytosis, secretion activation, cytotoxicity, and immunomodulation.
Expressed by B cells, monocytes, granulocytes, and platelets.
Involved in immune complex phagocytosis and antibody production regulation.
Certain CD32 variants may increase susceptibility to systemic lupus erythematosus (SLE).
