Product Specific Information

• The 93 monoclonal antibody reacts with an epitope shared by mouse CD16 and CD32.
• CD16 (FcγRIII) and CD32 (FcγRII) are low-affinity receptors for the mouse IgG Fc portion, expressed by B cells, monocytes/macrophages, NK cells, and neutrophils.
• Each product includes 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.
• Reported for use in flow cytometric analysis.
• Tested by flow cytometric analysis of mouse splenocytes at ≤1.0 µg/test (100 µL final volume).
• Recommended to titrate antibody for optimal performance.
• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining. Add 5 µL per sample prior to combining with cells.
• For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells).
• NovaFluor conjugates utilize Phiton™ technology, providing engineered fluorescent signatures optimized for spillover and spread.
• Excitation: 639 nm; Emission: 700 nm; Laser: 633–640 nm (Red).

Target Information

• CD16: 50–65 kDa cell surface molecule present in two forms — a transmembrane form (NK and some T cells) and a GPI-linked form (granulocytes). Functions as a low-affinity IgG receptor mediating ADCC.
• CD32: 40 kDa glycoprotein acting as a low-affinity receptor for IgG (FcγRII), mediating endocytosis, secretion activation, cytotoxicity, and immunomodulation.
• Expressed by B cells, monocytes, granulocytes, and platelets.
• Involved in immune complex phagocytosis and regulation of antibody production by B cells.
• Variants of CD32 may increase susceptibility to systemic lupus erythematosus (SLE).
• Several transcript variants encoding different isoforms of CD32 have been identified.
