Target Information
Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family, essential for T cell signal transduction, differentiation, and activation. CD8 is mainly expressed on T cells as a disulfide-linked heterodimer (CD8α/CD8β) acting as a co-receptor with TCR for MHC-I molecules, while CD4 interacts with MHC-II. CD8α is crucial for MHC-I binding and is also found on subsets of T cells, NK cells, monocytes, and dendritic cells as CD8α homodimers. Engagement of MHC-I/peptide complexes on antigen-presenting cells recruits Lck kinase, promoting lymphokine production, motility, and CTL activation. Activated CTLs are vital for pathogen and tumor cell clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
