Target Information

Huntington's disease (HD) is a neurodegenerative disorder characterized by loss of striatal neurons, caused by expansion of a polyglutamine tract in the huntingtin protein. HAP1 was identified via two-hybrid screening; its binding to huntingtin correlates with polyglutamine tract expansion.
HAP1 interacts with cytoskeletal proteins dynactin and pericentriolar autoantigen protein 1, suggesting roles in vesicular trafficking or organelle transport. It also participates in huntingtin-enhanced BDNF transport along microtubules. Disruption of this process leads to reduced neurotrophic support and neuronal toxicity, potentially contributing to disease pathogenesis. Several alternatively spliced isoforms of HAP1 have been described.
