Target Information
Presenilin 1 is a marker of susceptibility to early-onset Alzheimer’s disease. Together with PEN2, nicastrin, and APH-1, Presenilin 1 forms the γ-secretase protein complex—a membrane-bound aspartyl protease that cleaves certain proteins within the lipid bilayer. This cleavage is crucial for signaling from various cell-surface receptors and is implicated in generating neurotoxic amyloid peptides central to Alzheimer’s disease pathogenesis. γ-secretase cleaves amyloid precursor protein (APP) within its intramembrane region, producing either non-toxic p3 or toxic Abeta amyloid peptides. Accumulation of Abeta peptide is considered a precursor to Alzheimer’s disease. Multiple isoforms of Presenilin 1 are known to exist.
