• Reconstitution: Briefly centrifuge vial before opening. Reconstitute lyophilized recombinant human IL-21 in sterile distilled water to 0.1–1.0 mg/mL. Further dilutions should be made in low endotoxin medium or buffered solution containing a carrier protein (e.g., heat-inactivated FCS or tissue culture grade BSA).
• Storage: Store lyophilized recombinant human IL-21 at 2–8°C, preferably desiccated. After reconstitution, aliquot and store at ≤–20°C. Avoid repeated freeze/thaw cycles.

Target Information

IL-21 is a 17 kDa immunomodulatory cytokine mainly produced by NKT, Th17, and TFH cells. In TFH cells, IL-21 signaling through IL-21R and STAT3 activates Bcl6, essential for TFH cell development and function. IL-21 promotes T cell-dependent B cell differentiation into plasma and memory cells, stimulates IgG production, and induces apoptosis in naive B cells.
In Th17 cells, IL-21 enhances IL-23R expression via STAT3, IRF4, and RORγt, supporting maturation and maintenance by IL-23. It also downregulates Foxp3, influencing immune balance. IL-21 deficiency protects mice from chemically-induced colitis, while elevated IL-21 levels are observed in colitis models.
