Description:
This 4H9 monoclonal antibody reacts with human CD7 (Leu-9 or gp40), a 40-kDa transmembrane glycoprotein belonging to the immunoglobulin superfamily. CD7 is expressed on peripheral blood T lymphocytes and natural killer cells, as well as on lymphoid and myeloid lineage precursors during hematopoiesis. It is involved in T cell activation through association with CD3 and CD45.

CD7 expression is elevated on CD4+ T cells in classical Hodgkin’s lymphoma compared to benign reactive lymphadenopathy, making it useful as a diagnostic marker. Blocking assays show that this clone recognizes the same epitope as clone 124-1D1.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometric analysis on normal human peripheral blood cells. Use at 5 µL (0.25 µg) per test (100 µL final volume).
Excitation: 633–647 nm; Emission: 660 nm; Laser: Red Laser.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

CD7 (gp40, Leu9) is a 40 kDa member of the immunoglobulin gene superfamily. The N-terminal region (amino acids 1–107) is homologous to Ig kappa-L chains, while the C-terminal extracellular domain is proline-rich, forming a stalk-like structure. CD7 is expressed on most immature and mature T lymphocytes, T cell leukemias, NK cells, and some B cell populations. Cross-linking CD7 enhances T cell and NK cell activity, influencing calcium fluxes, adhesion molecule expression, cytokine secretion, and proliferation. It associates with phosphoinositol 3-kinase, inducing D-3 phosphoinositide production and tyrosine phosphorylation. CD7 expression serves as a marker in leukemia diagnostics.
