Product Specific Information

The B33B4WP monoclonal antibody recognizes human NFκB p65 subunit when serine 529 is phosphorylated.
NFκB is a transcription factor regulating genes involved in proliferation, apoptosis, immunity, and cancer.
Functional NFκB complexes are composed of p65 (RelA), c-Rel, RelB, p50, and p52 subunits.
Activation by TNFα, IL-1β, LPS, camptothecin, or PMA induces phosphorylation and degradation of IκB, allowing NFκB translocation to the nucleus.
Phosphorylation at Ser529 and/or Ser536 positively regulates p65 activity.

This antibody detects a single band at 65 kDa in TNFα-stimulated HeLa cell lysates.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated for use at 5 µL (0.06 µg)/test. Suitable for 10⁵–10⁸ cells/test.

Recommended Protocols:

• Protocol A: Two-step (cytoplasmic)
• Protocol C: Two-step (Fixation/Methanol)
• Protocol B (nuclear proteins): Not suitable

Target Information

Nuclear factor kB p65 (NF-kB p65), encoded by the RELA gene on chromosome 11, is also known as RelA.
It forms heterodimers with p50 or p52 to regulate transcriptional activity.
TNFα binding induces IKK-mediated phosphorylation of IκB, leading to IκB degradation and NFκB activation.
NFκB p65 plays a key role in transcription of immunoglobulin kappa gene in mature B-lymphoid cells.
