The CR55T33 monoclonal antibody recognizes phosphorylated serine 139 of human and mouse H2AX. H2AX is a member of the H2A histone family that complexes with DNA and other histones to form nucleosome units of eukaryotic chromatin.

After DNA damage such as double-strand breaks, genotoxic stress, or replication errors, PI3K-like kinases (ATM, ATR, DNA-PK) phosphorylate serine 139 in H2AX. This phosphorylation event recruits DNA repair proteins.

The CR55T33 antibody detects a single band (~15 kDa) on reduced lysates from etoposide-stimulated Jurkat cells.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated for intracellular staining using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00).
Recommended use: 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Target Information

Histone H2A.X (H2AX) is a core histone protein involved in nucleosome formation. Upon DNA double-strand breaks, serine 139 phosphorylation generates γ-H2AX, a marker of DNA damage.

Phosphorylation of H2AX can be detected via Western blot or immunofluorescence, reflecting DSB frequency. PI3K-family kinases (ATM, ATR, DNA-PK) are implicated in this process.

H2A.X contains 143 amino acids and exists in two mRNA forms (~1600 bases with polyA, ~575 bases without). It maps to chromosome 11q23.2–q23.3 and contributes to chromatin structure and DNA repair signaling.
