Product Specific Information
The MBM7C7 monoclonal antibody reacts with human and mouse BATF (B cell activating transcription factor). BATF is a 14 kDa member of the AP-1 family of transcription factors involved in the repression of other AP-1 family members. It is upregulated in activated T cells and is critical for Th17 differentiation, interacting with promoters of IL17, IL17F, IL21, and IL22. BATF is also implicated in CD8 T cell exhaustion in HIV infection.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and validated for intracellular staining and flow cytometric analysis of stimulated human peripheral blood cells using the Foxp3/Transcription Factor Buffer Set (Product # 00-5523-00).
Refer to BestProtocols® for staining protocol (Protocol B: One-step protocol for intracellular (nuclear) proteins).
Recommended use: 5 µL (0.25 µg) per test, in 100 µL final volume. Cell number: 10⁵–10⁸ cells/test (empirical determination required).

Target Information
BATF is a nuclear basic leucine zipper protein in the AP-1/ATF superfamily. Its leucine zipper mediates dimerization with Jun family members. BATF acts as a negative regulator of AP-1/ATF transcriptional events and inhibits Ras/Fos-mediated transformation. It is essential for differentiation of IL-17-producing T helper (Th17) cells and for proper class-switch recombination in T and B cells.
