Target Information
The c-Myc protein is a 62 kDa transcription factor encoded by the c-Myc gene on human chromosome 8q24. It is frequently activated in tumor cells and plays crucial roles in cellular proliferation, differentiation, apoptosis, and cell cycle progression. Phosphorylation at Thr58/Ser62 by kinases such as GSK3, CDK, ERK2, and JNK is associated with its regulation.
c-Myc is essential for vasculogenesis and angiogenesis during tumor development. Deregulated expression of c-Myc induces apoptosis and contributes to neoplastic transformation. Antibodies against c-Myc epitopes recognize overexpressed Myc-tagged proteins fused to amino- or carboxy-termini of target proteins.
Amplification of the c-Myc gene is observed in various human tumors, including lung, breast, and colon carcinomas. N-Myc amplification is typical in neuroblastomas, while c-Myc translocation to immunoglobulin loci (chromosomes 14, 2, or 22) is characteristic of Burkitt’s lymphoma and other B-cell lymphoproliferative disorders. Aberrant c-Myc expression occurs in tumors of multiple origins such as colorectal, gastric, hepatic, mammary, ovarian, and hematopoietic neoplasms.
