Product Specific Information

The monoclonal antibody TWAJ recognizes mouse and human Gata-3, a transcription factor critical for thymic development and Th2 differentiation. Expression occurs in the central nervous system, skin, mammary glands, and kidney during embryonic development. Homozygous knockout of Gata-3 is embryonic lethal.

Gata-3 is essential for T cell commitment and survival, primarily expressed in CD4 single-positive thymocytes. During Th2 differentiation, Gata-3 binds to the IL-4 promoter and represses T-bet expression, inhibiting Th1 differentiation.

Alternative splice variants (50 and 45 kDa) are recognized by this antibody.
Staining requires the Foxp3/Transcription Factor Staining Buffer Set (Product # 00-5523-00).
Cross-reactivity in rhesus monkeys has been documented.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-diluted and validated for intracellular staining of mouse thymocytes using the Foxp3/Transcription Factor Staining Buffer Set and protocol. Use at 5 µL (0.5 µg) per test (100 µL final volume, 10^5–10^8 cells/test).

Fluorochrome Information:
Brilliant Ultra Violet™ 661 (BUV661) emits at 660 nm and is optimized for UV (355 nm) laser-equipped cytometers. Ensure instrument compatibility.

Staining Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to reduce non-specific interactions.

Handling Notes:

  • Light sensitive – protect from light.
  • Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark.

Target Information

GATA3 binds enhancer elements of all four T-cell receptor genes (α, β, γ, δ), acting as an enhancer-binding protein with zinc finger domains. GATA3 mRNA is expressed in T cells but not in B cells or macrophages, contributing to TCR gene activation. The human GATA3 gene consists of 6 exons spanning 17 kb, with zinc finger domains encoded by separate exons highly conserved with GATA1.
