Product Specific Information
The J43 monoclonal antibody reacts with mouse PD-1 (programmed death-1), a 55 kDa Ig superfamily member involved in peripheral tolerance and autoimmune disease regulation. PD-1 is expressed mainly on activated T and B lymphocytes and interacts with PD-L1 (B7-H1) and PD-L2 (B7-DC).
J43 inhibits PD-L1-Ig and PD-L2-Ig binding to PD-1/BHK cells and modulates immune responses in vivo.
This antibody is validated for flow cytometric analysis of mouse splenocytes and can be used at ≤0.5 µg/test (100 µL final volume). Optimize cell number empirically (10⁵–10⁸ cells/test).
Fluorochrome Information:
Brilliant Ultra Violet™ 661 (BUV661) emits at 660 nm and is compatible with UV (355 nm) laser-equipped cytometers. Protect from light due to photo-induced oxidation sensitivity.
Staining Buffer Recommendation:
Use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to minimize non-specific polymer interactions when using multiple Brilliant dyes.
Fixation:
Samples can be stored up to 3 days at 4°C in IC Fixation Buffer (00-8222-49) or 1-step Fix/Lyse Solution (00-5333-54) with minimal fluorescence loss.

Target Information
T-cell activation is regulated by PD-1 (PDCD-1), which binds ligands PD-L1 and PD-L2 to deliver inhibitory signals from antigen-presenting cells, maintaining immune tolerance. PD-1 deficiency leads to hyperactive immune responses and autoimmune phenotypes in mice.
Although PD-1 has a predicted molecular weight of 55 kDa, it often migrates at a higher molecular weight in SDS-PAGE.
