Description:
The monoclonal antibody CXCR3-173 recognizes mouse CD183 (CXCR3), a seven-transmembrane G-protein coupled chemokine receptor that binds CXCL9 (Mig), CXCL10 (IP-10), and CXCL11 (ITAC). CD183 plays a role in CD4 T cell responses to grafts, with knockout mice showing compromised allograft rejection. Expression is found on NK cells, subsets of T lymphocytes, Tregs, and Th1-polarized cells. The antibody affects chemotaxis in response to ligand, and ligand presence eliminates staining. In vivo, antibody addition delays cardiac and pancreatic islet allograft rejection.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported:

• Flow cytometric analysis of mouse splenocytes.

Applications Tested:

• Tested for flow cytometry at ≤0.25 µg/test (100 µL final volume).
• Recommended to titrate for optimal performance.
• Cell number range: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

This gene encodes a G protein-coupled receptor selective for chemokines IP10, Mig, and I-TAC. These chemokines belong to the CXC subfamily, characterized by conserved cysteine residues. Binding induces leukocyte trafficking responses such as integrin activation, cytoskeletal changes, and chemotactic migration. The receptor couples with Gi-class G proteins, and signaling may involve pathways similar to other chemokine receptors. Expression is prominent in effector/memory T cells and T cells in inflamed tissues. IP10, Mig, and I-TAC are produced in inflammatory lesions, suggesting involvement in immune cell recruitment. The receptor is a potential therapeutic target for inflammatory diseases.
