Product Specific Information
The monoclonal antibody CXCR3-173 recognizes mouse CD183 (CXCR3), a seven-transmembrane G-protein-linked chemokine receptor binding CXCL9 (Mig), CXCL10 (IP-10), and CXCL11 (ITAC). CD183 plays a role in CD4 T cell responses to grafts, and CXCR3 knockout mice show compromised allograft rejection. Expression is found on NK cells, subsets of T lymphocytes and Tregs, and preferentially on Th1-polarized cells. CXCR3-173 affects chemotaxis in response to ligand; ligand presence eliminates antibody staining. In vivo, antibody addition delays cardiac and pancreatic islet allograft rejection.

Applications Tested: Tested on mouse splenocytes at ≤0.25 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Antibody should be titrated for optimal performance.
NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for best results.
CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates. Add 5 µL per sample to antibody cocktails/master mixes prior to combining with cells. For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).
NovaFluor conjugates utilize Phiton™ technology, based on engineered nucleic acid dye structures for optimized fluorescent signatures minimizing spillover and spread.

Target Information
This gene encodes a G protein-coupled receptor selective for chemokines IP10, Mig, and I-TAC, part of the CXC chemokine family. Binding induces leukocyte traffic responses such as integrin activation, cytoskeletal changes, and chemotactic migration. Expression is prominent in effector/memory T cells and inflammatory tissues. This receptor is a potential target for small-molecule antagonists for inflammatory disease treatment. Multiple transcript variants exist.
