Product Specific Information

The MIH5 monoclonal antibody reacts with mouse B7-H1 (PD-L1), a member of the B7 family (~40 kDa, Ig superfamily). PD-L1 is expressed on most leukocytes including T, B, NK, and dendritic cells and serves as a ligand for PD-1, leading to inhibition of T and B cell responses. MIH5 is reported to be a blocking antibody.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications

• Reported for use in flow cytometric analysis of mouse splenocytes.
• Recommended to titrate antibody for optimal performance.
• Use ≤0.06 µg per test in 100 µL final volume.

Important Notes

• NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, or DAPI.
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
  • Add 5 µL per sample before adding cells.
  • For single-color controls, add 5 µL per 100 µL cell sample (10³–10⁸ cells).

Internal testing shows strain-dependent staining differences: NovaFluor Yellow 700 may non-specifically stain B cells in SJL mice, but not in BALB/c or C57BL/6 strains.

Target Information

Programmed death receptor ligand 1 (PD-L1, B7-H1) is a B7 family member that binds to PD-1, negatively regulating T-cell receptor (TCR) signaling. PD-L1 engagement reduces TCR-mediated proliferation and cytokine production. PD-L1 is abundantly expressed on murine and human cancers and upregulated by IFN-γ, suggesting a key role in tumor immune evasion.
