Product Specific Information
The monoclonal antibody CXCR3-173 recognizes mouse CD183 (CXCR3), a seven-transmembrane G-protein linked chemokine receptor binding CXCL9 (Mig), CXCL10 (IP-10), and CXCL11 (ITAC).
CD183 plays a role in CD4 T cell responses to grafts; knockout mice show compromised allograft rejection. Expression is found on NK cells, subsets of T lymphocytes and Tregs, and Th1-polarized cells. The antibody affects chemotaxis in response to ligand, and ligand presence eliminates staining. In vivo addition delays cardiac and pancreatic islet allograft rejection.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤ 0.25 µg/test (100 µL final volume). Optimize concentration empirically for best performance.
Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (PI, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
- CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
- Add 5 µL per sample to antibody cocktails/master mixes before combining with cells.
- For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).
NovaFluor conjugates utilize Phiton™ technology, providing engineered fluorescent signatures optimized for spillover and spread impacts.

Target Information
This gene encodes a G protein-coupled receptor selective for chemokines IP10, Mig, and I-TAC. These belong to the CXC chemokine family, inducing leukocyte traffic via integrin activation and chemotactic migration.
Inhibition by Bordetella pertussis toxin suggests coupling with Gi-class G protein. Responses are rapid and transient, restored after receptor recycling.
Prominently expressed in effector/memory T cells and inflamed tissues, contributing to inflammatory cell recruitment.
Target for small molecular antagonists in inflammatory disease research. Multiple transcript variants encoding different isoforms exist.
