Product Specific Information

The monoclonal antibody CXCR3-173 recognizes mouse CD183 (CXCR3), a seven-transmembrane G-protein-coupled chemokine receptor that binds CXCL9 (Mig), CXCL10 (IP-10), and CXCL11 (ITAC). CD183 is involved in CD4 T cell responses to grafts, and knockout mice show compromised allograft rejection responses. Expression is found on NK cells, subsets of T lymphocytes, Tregs, and preferentially on Th1-polarized cells.

The antibody affects chemotaxis in response to ligand; ligand presence eliminates staining. In vivo, antibody addition delays cardiac and pancreatic islet allograft rejection.

A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number may range from 10⁵ to 10⁸ cells/test. Antibody titration is recommended for optimal performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
• Add 5 µL per sample to antibody cocktails or single-color controls.

Target Information

This gene encodes a G protein-coupled receptor selective for chemokines IP10, Mig, and I-TAC. These chemokines induce cellular responses involved in leukocyte trafficking, integrin activation, cytoskeletal changes, and chemotactic migration. The receptor is coupled to Gi-class G proteins and participates in rapid, transient signaling due to receptor internalization and recycling.

It is prominently expressed in effector/memory T cells and inflamed tissues, suggesting a role in inflammatory cell recruitment. This receptor is a potential target for small-molecule antagonists in inflammatory disease research.
