Product Specific Information

The monoclonal antibody Moushi101 recognizes mouse CD101 (IGSF2, V7), a transmembrane glycoprotein with 7 IgV domains and a short cytoplasmic tail containing phosphorylation sites.
CD101 cross-linking can antagonize TCR/CD3-mediated lymphocyte activation.
CD101 is expressed on monocytes, granulocytes, dendritic cells, Langerhans cells, and activated T cells, as well as on a subset of CD25+Foxp3+ T regulatory cells with high suppressive activity.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse splenocytes, ≤0.25 µg/test (100 µL final volume, 10⁵–10⁸ cells/test).
Careful titration recommended for optimal performance.

Compatibility Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):
Required for optimal staining with NovaFluor conjugates.
Add 5 µL per sample (100 µL cell sample, 10³–10⁸ cells) before combining with cells.

NovaFluor conjugates utilize Phiton™ technology, providing engineered fluorescent signatures optimized for minimal spillover and spread.

Target Information

CD101 (V7 antigen) is a member of the immunoglobulin superfamily and forms a disulfide-linked homodimer.
It is expressed on monocytes, granulocytes, dendritic cells (Langerhans-like), and activated T lymphocytes.
CD101+ CD28+ cells respond strongly to CD28 signaling and can enhance proliferation when combined with anti-CD28 or suboptimal anti-CD3 stimulation.
Mouse CD101 is found on CD4+CD25+Foxp3+ regulatory T cells with high suppressor activity.
In human PBMCs, CD101 is expressed on ~30% of Foxp3+ cells, though suppressive function is less defined.
