Product Specific Information
The monoclonal antibody Moushi101 recognizes mouse CD101, also known as IGSF2 and V7 (7 IgV domains). CD101 is a transmembrane glycoprotein with 7 Ig-like domains and a short cytoplasmic tail containing phosphorylation sites. Cross-linking CD101 can antagonize TCR/CD3-mediated lymphocyte activation. CD101 is expressed on monocytes, granulocytes, dendritic cells, Langerhans cells, and activated T cells. It is also found on a subset of CD25⁺Foxp3⁺ regulatory T cells with high suppressive properties.

Applications Tested: Tested on mouse splenocytes; ≤0.25 µg/test recommended.
A test is defined as the antibody amount required to stain cells in 100 µL volume. Cell count may range from 10⁵–10⁸ cells/test. Optimize dilution for best performance.
Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
- NovaFluor conjugates ship with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which improves staining performance.
- Add 5 µL of CellBlox Plus per sample (regardless of the number of NovaFluors).
- For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).
NovaFluor conjugates utilize Phiton™ technology, employing nucleic acid dye structures to minimize spectral spillover and spread effects.

Internal testing indicates NovaFluor Yellow 730 may non-specifically stain B cells in SJL mice, but not in BALB/c or C57BL/6 strains.
Target Information
CD101 (V7 antigen) is a disulfide-linked homodimeric member of the immunoglobulin superfamily. It is expressed on monocytes, granulocytes, dendritic cells (Langerhans-like), and T lymphocytes. CD101⁺CD28⁺ cells respond strongly to CD28 signaling and can enhance proliferation when co-stimulated with anti-CD28 or suboptimal anti-CD3, suggesting an activating role.
In mice, CD101 is present on a subpopulation of regulatory T cells (CD4⁺, CD25⁺, Foxp3⁺) with high suppressor activity. In human PBMCs, approximately 30% of Foxp3⁺ cells express CD101, although increased suppressor activity has not been confirmed.
