Background
Saliva is a non-invasive alternative to blood collection for genomic DNA extraction. DNA from buccal epithelial cells and leukocytes in saliva can be used for disease diagnosis, monitoring, and infection detection.
Norgen’s Saliva DNA Isolation Kit enables fast and reliable DNA isolation from both preserved and fresh saliva samples.

Total DNA was isolated from 250 µL of eight fresh saliva samples using Norgen’s kit.
For evaluation, 10 µL of each 100 µL elution was run on a 1.2% agarose gel.
High yield and quality were observed in all lanes.
Lane M: Norgen’s FastRunner 1 kb DNA Ladder.

Q. What to do if column gets clogged?
Ensure proper centrifugation speed/time, apply ≤0.5 mL saliva, and vortex lysate before loading.
Q. Why low DNA yield?
May result from incomplete lysis or low DNA concentration in sample; extend Proteinase K incubation.
Q. Poor downstream performance?
Ensure thorough washing and removal of ethanol before elution.
Q. RNA contamination?
Perform RNase A digestion if RNA interferes with downstream use.
