Target Information

Amyloid beta peptide (Abeta/Beta-amyloid) is the major constituent of amyloid plaques in the brains of individuals afflicted with Alzheimer’s disease.
Abeta peptide (40–43 amino acids) is generated from the beta-amyloid precursor protein (βAPP) through a two-step proteolytic process involving β-secretase (BACE) and γ-secretase.
The cleavage by BACE releases a soluble fragment and leaves a membrane-bound remnant, which is further processed by γ-secretase to produce amyloid beta peptide.
γ-secretase is a multi-subunit complex containing presenilin-1 and -2, and associated with nicastrin, which modulates Abeta production.
Abeta forms extracellular filamentous deposits found in amyloid plaques and neurofibrillary tangles, characteristic of Alzheimer’s disease and other amyloid-related disorders such as Lewy body dementia, Down’s syndrome, and cerebral amyloid angiopathy.
