Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human sFas Receptor. Anti-Human sFas Receptor-specific antibody was purified by affinity chromatography employing an immobilized Human sFas Receptor matrix.

Sandwich ELISA:
To detect Human sFas Receptor by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.5–2.0 µg/mL of this antibody is required. This antigen affinity purified antibody, in conjunction with PeproTech Biotinylated Anti-Human sFas Receptor (500-P295Bt) as a detection antibody, allows detection of at least 0.2–0.4 ng/well of Recombinant Human sFas Receptor.

Western Blot:
To detect Human sFas Receptor by Western Blot analysis, this antibody can be used at a concentration of 0.1–0.2 µg/mL. When used with compatible secondary reagents, the detection limit for Recombinant Human sFas Receptor is 1.5–3.0 ng/lane, under either reducing or non-reducing conditions.

Target Information

FAS (Fas, CD95, APO-1) is a 46 kDa transmembrane glycoprotein functioning as a cell death receptor in the TNFR (tumor necrosis factor receptor) superfamily. It also exists in a soluble form (~26 kDa). Stimulation of FAS leads to aggregation of intracellular FAS-associated death domains (FADD), formation of the death-inducing signaling complex (DISC), and activation of caspases, triggering apoptosis.
FAS has been shown to activate NF-kappaB, MAPK3/ERK1, MAPK8/JNK, and is involved in proliferative signaling in fibroblast and T cells. Multiple splice variants encode distinct isoforms; soluble isoforms may negatively regulate apoptosis. The Fas/Fas ligand system plays roles in diseases such as AIDS, hepatitis, and cancer, and is key in antiviral immune responses.
