Target Information
Spleen tyrosine kinase (SYK) is a cytoplasmic tyrosine kinase associated with allergic and autoimmune disease pathways, and may also play a role in cancer. Upon activation of B cell antigen receptor (BCR) or high-affinity IgE receptor (FcεRI), SYK translocates to the plasma membrane and phosphorylates adaptor proteins, initiating downstream signaling such as calcium mobilization, cytoskeleton remodeling, and gene transcription. SYK interacts with receptor assemblies via SH2 domain binding to ITAM motifs phosphorylated by Src-family kinases, which also contribute to SYK activation.
