Target Information
Spleen tyrosine kinase (SYK), along with ZAP70, are cytoplasmic tyrosine kinases that are potential drug targets for allergic disorders and autoimmune diseases. SYK may also play a role in cancer. Upon activation of B cell antigen receptor (BCR) or high-affinity IgE receptor (FcεRI), Syk translocates to the plasma membrane and phosphorylates downstream adaptor proteins, initiating signaling pathways such as calcium mobilization, cytoskeleton remodeling, and gene transcription. Syk binds to receptor assemblies via SH2 domain interactions with phosphorylated ITAM motifs, which are modified by Src-family kinases that also activate Syk through phosphorylation.
