Target Information

MDM2 is a ubiquitin ligase for p53 and plays a central role in regulation of p53 stability. Located on chromosome 12q, MDM2 is phosphorylated by Akt at Ser166 and Ser186, enhancing its interaction with p300 and promoting p53 ubiquitination and degradation. Phosphorylation also inhibits MDM2 binding to p19ARF, further increasing p53 degradation. MDM2 negatively regulates p53 by inhibiting its transactivation and promoting its degradation via E3 ubiquitin ligase activity. Overexpression of MDM2 leads to p53 inactivation and reduced tumor suppressor function. MDM2 is implicated in apoptosis, cell cycle regulation, and is overexpressed in various malignancies including soft tissue carcinomas and breast cancer. It interacts with proteins such as retinoblastoma 1 and ribosomal protein L5, and more than 40 alternatively spliced variants have been identified in both tumor and normal tissues.
