Antibody detects endogenous levels of Tau only when phosphorylated at Ser739, which site historically referenced as Ser422.


Target Information

Tau is a neuronal microtubule-associated protein found predominantly on axons. It promotes tubulin polymerization and stabilizes microtubules.
The C-terminus binds axonal microtubules while the N-terminus binds neural plasma membrane components, functioning as a linker between both.
Axonal polarity is predetermined by TAU/MAPT localization in the neuronal cell body domain defined by the centrosome.
Short isoforms allow cytoskeletal plasticity, while longer isoforms may play a role in stabilization.
In its hyper-phosphorylated form, Tau is the major component of paired helical filaments (PHF), building blocks of neurofibrillary lesions in Alzheimer’s disease brain.
Hyper-phosphorylation impairs Tau’s microtubule binding, destabilizing microtubules and leading to neuronal degeneration.
Numerous serine/threonine kinases phosphorylate Tau, including GSK-3β, PKA, cdk5, and casein kinase II.
Hyper-phosphorylated Tau is found in neurofibrillary lesions in Alzheimer’s disease and other CNS disorders such as Pick’s disease, frontotemporal dementia, corticobasal degeneration, and progressive supranuclear palsy.
