Description:
This affinity-purified polyclonal rabbit antibody reacts with mouse and rat tumor necrosis factor-alpha (TNF-α). Mouse TNF-α is a 17 kDa factor produced by macrophages, monocytes, neutrophils, CD4 T cells, and NK cells. A 26 kDa membrane-bound form also exists. TNF-α is cytolytic and crucial for immune regulation, existing as a bioactive homotrimer.

Applications Reported:
This antibody has been reported for use as the plate-coating (ELISA Capture) antibody in the In Vivo Cytokine Capture Assay (IVCCA), used with the Functional Grade Biotinylated TN3.19 antibody for detection. The IVCCA method enhances cytokine detection sensitivity by extending in vivo half-life up to 1000-fold.

Applications Tested:
Tested as the ELISA capture antibody in a sandwich ELISA for mouse TNF-α analysis, used with Functional Grade Biotinylated anti-mouse TNF-α antibody (14-8321) for detection and recombinant mouse TNF-α as the standard.
Recommended concentration for capture: 1.0–4.0 µg/mL

Target Information

TNF-α is a multifunctional proinflammatory cytokine belonging to the TNF superfamily. It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF-α regulates immune cells, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. It appears as a 17 kDa band under non-reducing SDS-PAGE conditions.
Knockout studies suggest TNF-α’s neuroprotective role and its ability to induce tumor necrosis in vivo. It is involved in immune responses to bacterial, viral, parasitic, and fungal infections, as well as tumor necrosis. TNF-α acts synergistically with interferon-gamma in cytotoxicity and is implicated in autoimmune diseases, insulin resistance, and cancer.
