Description:
The MAb11 antibody reacts with human tumor necrosis factor-alpha (TNF alpha), a 17 kDa cytokine produced by monocytes, macrophages, neutrophils, NK cells, and CD4+ T cells. TNF alpha exhibits cytolytic activity against a range of tumor cells and plays an important role in immune regulation. It exists as dimers and trimers and also as a 26 kDa membrane-bound form. MAb11 is a neutralizing antibody.

Applications Reported:

• Detection of human TNF alpha by ELISA
• Intracellular flow cytometric analysis

Applications Tested:
The biotinylated format of MAb11 has been tested in sandwich ELISA for detection of human TNF alpha in combination with MAb1 and can be used at 1 µg/mL. It is recommended to titrate the antibody for optimal assay performance.

Target Information

TNF alpha is a multifunctional proinflammatory cytokine belonging to the TNF superfamily. It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF alpha regulates immune cell activity, proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. It forms multimers (dimers, trimers, or pentamers) and appears as a 17 kDa band under non-reducing SDS-PAGE conditions.
Knockout studies in mice suggest TNF alpha has neuroprotective functions and induces tumor necrosis in tumor-bearing mice. TNF alpha is involved in immune responses to bacterial, viral, parasitic, and fungal infections, and contributes to tumor cytotoxicity. It has been implicated in autoimmune diseases, insulin resistance, and cancer.
