Description:
The MAb1 antibody reacts with human tumor necrosis factor-alpha (TNF-alpha), a 17 kDa cytokine produced by monocytes, macrophages, neutrophils, NK cells, and CD4⁺ T cells. TNF-alpha has cytolytic activity against various tumor cells and plays a crucial role in immune regulation. It forms dimers and trimers and also exists as a 26 kDa membrane-bound form.
Applications Reported:
MAb1 has been reported for use in ELISA capture of human TNF-alpha, immunoblotting (WB), and neutralization of TNF-alpha activity. For functional assays, use the Functional Grade purified MAb1 (Product #16-7348).
Applications Tested:
MAb1 was tested as the capture antibody in a sandwich ELISA with biotin MAb11 (13-7349) as detection and recombinant human TNF-alpha (14-8329) as the standard. Recommended ELISA capture range: 1–4 µg/mL.
A standard curve using recombinant TNF-alpha (1000 pg/mL–8 pg/mL) should be included in each ELISA plate.

Target Information
TNF-alpha is a multifunctional proinflammatory cytokine belonging to the TNF superfamily. It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR.
It regulates immune response, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. TNF-alpha exists as multimers (dimers, trimers, or pentamers) and appears as a single 17 kDa band under non-reducing SDS-PAGE conditions.
It plays roles in tumor necrosis, immune defense against pathogens, and has implications in autoimmune diseases, insulin resistance, and cancer.
