Product Specific Information

This 37D12 monoclonal antibody reacts with human IL-37, a member of the interleukin 1 cytokine family (also known as IL-1F7 or IL-1H4). IL-37 suppresses innate inflammatory and immune responses and exhibits anti-inflammatory characteristics similar to IL-1R8. It binds the alpha chain of IL-18R with low affinity and does not recruit IL-18 beta, thus not mediating pro-inflammatory activity. IL-37 also interacts with IL-18 binding protein (IL-18BP), enhancing its antagonistic effects and forming a trimeric complex with IL-18BP and IL-18R beta to block IL-18 activity.

Transcripts of IL-37 have been detected in human tissues including lung, testis, and colon tumors, as well as in several human cell lines.

Applications Reported: Suitable for immunocytochemistry followed by cytospin.
Applications Tested: Tested on fixed and permeabilized normal human peripheral blood mononuclear cells at ≤20 µg/mL. Optimal dilution should be titrated for specific assays.

Target Information

The IL-37 protein belongs to the interleukin 1 cytokine family. It can bind to interleukin 18 receptor (IL18R1/IL-1Rrp) and to interleukin 18 binding protein (IL18BP), forming a complex with IL18 receptor beta subunit that inhibits IL18 activity. This gene is part of a cytokine gene cluster on chromosome 2, and five alternatively spliced transcript variants encoding distinct isoforms have been reported.
