Product Specific Information

The monoclonal antibody AE1 recognizes many members of the acidic (Type I) family of cytokeratins. Cytokeratins are intermediate filament proteins forming part of the cytoskeleton. There are two large families of cytokeratins, acidic and basic, sharing common domains such as an alpha-helical core with N- and C-terminal regions. These proteins are expressed in epithelial cells and are developmentally regulated. Many tumors also express these cytokeratins, helping identify neoplasm origins.

The AE1 monoclonal antibody recognizes 56.5, 54’, 50, 50’, 48, and 40 kDa proteins (CK10, 14, 15, 16, 19) in mouse, human, rat, primate (cynomolgus and rhesus), dog, cat, rabbit, and chicken.

Applications Reported:
Suitable for western blotting, immunohistochemical staining of frozen or paraffin-embedded tissue sections, and immunocytochemistry.

Applications Tested:
Tested by western blot on SDS-reduced lysates from MCF7 cells at ≤2 µg/mL. It is recommended to titrate the antibody for optimal performance.

Purity: Greater than 90%, determined by SDS-PAGE.
Aggregation: Less than 10%, determined by HPLC.
Filtration: 0.2 µm post-manufacturing filtered.


Target Information

Cytokeratin pan belongs to a subfamily of intermediate filament proteins characterized by biochemical diversity, represented in human epithelial tissues by at least 20 different polypeptides. Cytokeratins range from 40–68 kDa with isoelectric points between 4.9–7.8.
Individual cytokeratins are numbered 1–20, and their expression patterns vary by epithelium type and differentiation stage. These patterns are valuable for distinguishing epithelial malignancies. Cytokeratin antibodies assist in tumor diagnosis via immunohistochemistry and are useful in cytopathology and flow cytometry assays.
Their composition changes depending on epithelial cell type, differentiation stage, and disease state, making keratins important markers in cancer research and tumor diagnostics.
