Product Specific Information
The ITEM-1 monoclonal antibody reacts with human TWEAK Receptor/Fn14 (fibroblast growth factor-inducible 14 kDa protein). Fn14 is distantly related to the TNFR family, containing one cysteine-rich domain and a TNFR-associated factor binding domain but lacking a death domain. Fn14 is involved in TWEAK-induced endothelial cell migration, proliferation, and angiogenesis. TWEAK-induced cell death via Fn14 includes both apoptosis and necrosis, and can be blocked by anti-TWEAK antibody CARL-1. Fn14 is expressed on HUVEC and certain cancer tissues but not on freshly isolated PBMCs. Fn14 mRNA expression has been identified during liver regeneration.
Applications Reported: ITEM-1 antibody has been used for flow cytometric analysis and immunohistochemical staining of frozen tissue sections. It has also been used in induction of cell death in transfected cells (use Functional Grade purified ITEM-1 for functional assays).
Applications Tested: Tested by flow cytometric analysis on human Fn14 transfected cells. Recommended ≤1 µg per test (100 µL final volume). Cell number should be determined empirically (10^5–10^8 cells/test). Antibody titration recommended for optimal performance.
Target Information
TWEAK is a member of the TNF family and acts as a weak inducer of apoptosis. Its receptor, TweakR (Fn14), was isolated from a HUVEC cDNA library and belongs to the TNF receptor superfamily. TweakR initiates proliferative signaling by binding to TRAF1, 2, and 3. TweakR mRNA is upregulated during tissue regeneration in the aorta and liver and may play roles in vascular cell migration and angiogenesis.
