Product Specific Information
The 6C2 monoclonal antibody reacts with mouse Toll-like receptor 2 (TLR2). Mouse TLR2 is expressed by myeloid lineage cells, including macrophages and dendritic cells in splenocytes and the RAW264.7 cell line. The Toll family includes at least twelve members in human and mouse, characterized by an extracellular leucine-rich repeat domain and a cytoplasmic domain homologous to the type I IL-1 receptor.
TLR2 and TLR4 act as pattern recognition receptors and signaling molecules in response to bacterial lipoproteins, playing key roles in innate immunity and inflammation. TLR2 is expressed on the cell surface and distinguishes between different pathogens.
Applications Reported:
This antibody is reported for use in flow cytometry, immunoprecipitation, and immunohistology of frozen sections. Preliminary studies indicate partial inhibition of monocyte activation through TLR2. For optimal flow cytometry, a directly conjugated 6C2 (FITC, PE, or Biotin) is recommended along with FcR blocking using anti-mouse CD16/CD32 (clone 93, Product #14-0161).
Applications Tested:
Tested in flow cytometry using mouse splenocytes or RAW264.7 cells. Recommended ≤1 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Titrate for optimal performance.

Target Information
TLR2 is a member of the Toll-like receptor family involved in pathogen recognition and activation of innate immunity. TLR2 signals through adaptor molecules such as MyD88 and TIRAP to activate kinases and transcription factors including PKCα/β and NF-κB.
It forms heterodimers with TLR1 or TLR6 to recognize bacterial and mycoplasma lipoproteins. TLR2 is expressed mainly in peripheral blood leukocytes and mediates responses to Gram-positive bacteria and yeast via NF-κB activation.
Human TLR2 is expressed in tonsils, microglia, lymph nodes, appendices, and activated B-cells. Expression is highest in CD14⁺ monocytes, followed by CD15⁺ granulocytes, CD19⁺ B-cells, and CD3⁺ T-cells. Expression is upregulated by LPS, GM-CSF, IL-1, and IL-10, and downregulated by IL-4, IFN-γ, and TNF.
