Product Specific Information

This 22F6 monoclonal antibody reacts with human, mouse, and canine Helios. Helios is a zinc-finger transcription factor similar to Ikaros, expressed in hematopoietic stem cells of embryos and adult mice. Highest expression is in regulatory T cells, but not in mature B cells, dendritic cells, or myeloid cells.
Helios can form homodimers or heterodimers with other Ikaros family members to regulate transcription. Eight isoforms exist, some with dominant-negative functions, upregulated in lymphoid malignancies such as T-acute lymphoblastic leukemia.
22F6 has been reported to cross-react with rat Helios.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometric analysis of mouse lymph node cells. Recommended at 5 µL (0.25 µg) per test (100 µL final volume). Cell count range: 10⁵–10⁸ cells/test.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be verified empirically.

Target Information

This gene encodes a member of the Ikaros family of zinc-finger proteins (Ikaros, Aiolos, Helios). These hematopoietic-specific transcription factors regulate lymphocyte development. Helios forms homo- or hetero-dimers with other Ikaros family members and functions mainly in early hematopoietic development. Multiple transcript variants exist; biological validity of some variants remains uncertain.
