Product Specific Information
The monoclonal antibody 3E4 reacts with human and mouse interferon regulatory factor 4 (IRF4), a 52 kDa transcription factor involved in B cell, T cell, and macrophage function.
- In B cells, IRF4 is highly expressed in mature plasma cells and regulates differentiation and transcription of genes such as Prdm1 (Blimp1).
- In T cells, IRF4 contributes to Treg, Th2, Th9, and Th17 development and function, inducing IL-10 expression.
- IRF4 also plays a role in macrophage polarization and is upregulated in various hematologic malignancies.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Intracellular staining of stimulated human peripheral blood cells using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00). Recommended ≤0.25 µg/test in 100 µL final volume. Cell number: 10⁵–10⁸ cells/test. Optimize titration for best results.

Alexa Fluor 700 emits at 723 nm and is excited by red laser (633–647 nm). Use 685 LP mirror and 710/20 band pass filter.

Target Information
The IRF (interferon regulatory factor) family are transcription factors with a tryptophan pentad repeat DNA-binding domain, regulating interferon response and interferon-inducible genes.
IRF4 is lymphocyte-specific and negatively regulates TLR signaling, influencing innate and adaptive immunity. A chromosomal translocation t(6;14)(p25;q32) involving IRF4 and IgH may cause multiple myeloma. Alternative splicing variants exist.
