Product Specific Information
The HTA125 monoclonal antibody reacts with human Toll-like receptor 4 (TLR4). TLR4, a member of the Toll family, is a type I transmembrane protein with leucine-rich repeats and a cytoplasmic domain homologous to the IL-1 receptor. It forms a complex with MD-2 and CD14, mediating LPS recognition and signaling in innate immunity and inflammation. TLR4 is expressed on peripheral blood monocytes.
HTA125 immunoprecipitates human TLR4 (~100 kDa) from transfected cells. TLR surface expression varies among donors and is generally low on monocytes and other immune cells. For optimal staining, it is recommended to use the lysed whole blood protocol rather than Ficoll-gradient prepared cells, as the latter may reduce staining intensity.
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated for flow cytometry at 5 µL (1.0 µg) per test (100 µL final volume). Suitable for 10⁵–10⁸ cells/test.

Alexa Fluor™ 700 emits at 723 nm, excited by the red laser (633 nm). Recommended filter: 685 LP mirror, 710/20 filter.

Target Information
TLR4 (Toll-like receptor 4) plays a key role in pathogen recognition and activation of innate immunity. It is expressed in placenta and myelomonocytic leukocytes, responding to LPS via a receptor complex with CD14 and LY96. TLR4 mediates cytokine production essential for immune responses. Mutations in TLR4 influence LPS responsiveness. Multiple transcript variants exist, though their protein-coding potential is uncertain. TLR4-deficient mice show increased susceptibility to Gram-negative bacterial infection.
