Target Information
CD22 (BL-CAM) is a type I integral membrane glycoprotein with a molecular weight of 130–140 kDa, expressed in both the cytoplasm and cell membrane of B-lymphocytes.
It appears early in B-cell differentiation, similar to CD19, and is expressed in mature B cells (CD22⁺) but not in plasma cells (CD22⁻).
CD22 is strongly expressed in hairy cell leukemia and may be useful in phenotyping mature leukemias.
It preferentially binds to α2,6-linked sialic acid, and its recognition site can be masked by cis interactions on the same cell surface.
Upon ligand-induced tyrosine phosphorylation, CD22 participates in regulating B-cell receptor signaling.
It can act as both a positive regulator through interaction with Src family kinases and as an inhibitory receptor by recruiting SH2-domain phosphatases that dephosphorylate signaling molecules.
CD22 is positive in diffuse large B-cell lymphoma and nodular lymphocyte predominance Hodgkin’s lymphoma but negative in classical Hodgkin’s lymphoma.
