Target Information
Adducins are heteromeric proteins composed of alpha, beta, and gamma subunits, encoded by distinct genes. They are membrane skeletal proteins involved in the assembly of the spectrin–actin network in erythrocytes and at cell–cell contact sites in epithelial tissues.
- Alpha and gamma subunits are ubiquitously expressed.
- Beta subunit expression is restricted to brain and hematopoietic tissues.
- Adducin was originally purified from human erythrocytes as a heterodimer of alpha and beta subunits.
- Polymorphisms in alpha and beta subunits are linked to blood pressure regulation in animal models of hypertension.
- Heterodimers of alpha and gamma subunits also exist.
Each subunit has two domains: - Amino-terminal region: protease-resistant and globular.
- Carboxy-terminal region: protease-sensitive, contains phosphorylation sites for protein kinase C, calmodulin-binding site, and regions required for spectrin and actin association.
Alternative splicing of adducin gamma transcripts produces different isoforms, though their functions remain unclear.
