Target Information

Adducins are heteromeric proteins composed of alpha, beta, and gamma subunits, encoded by distinct genes. They are membrane skeletal proteins involved in assembling the spectrin–actin network in erythrocytes and epithelial tissues.

• Alpha and gamma subunits are ubiquitously expressed, while beta is restricted to brain and hematopoietic tissues.
• Adducin was first purified from human erythrocytes as a heterodimer of alpha and beta subunits.
• Polymorphisms in these subunits have been linked to blood pressure regulation in animal models of hypertension.
• Heterodimers of alpha and gamma subunits also exist.
• Each subunit has two domains: a protease-resistant globular N-terminal region and a protease-sensitive C-terminal region containing phosphorylation sites for protein kinase C, a calmodulin-binding site, and regions required for spectrin and actin association.
• Alternatively spliced gamma isoforms exist, but their functions remain unclear.
