Target Information

Proteolytic degradation is critical to maintaining appropriate levels of short-lived and regulatory proteins involved in diverse cellular processes such as metabolism, stress response, antigen presentation, receptor modulation, ion channel regulation, cell cycle, transcription, and signaling.
The ubiquitin-proteasome pathway deconstructs most cytosolic and nuclear proteins in eukaryotic cells, while others are degraded via the vacuolar pathway, including endosomes, lysosomes, and the endoplasmic reticulum.
The 26S proteasome is an ATP-dependent, multisubunit complex (~2.5 MDa) consisting of a 20S proteolytic core and 19S regulatory subunits that recognize ubiquitinated proteins, unfold them, and translocate them into the 20S core.
Defects in the ubiquitin-proteasome pathway are linked to several genetic diseases, including cystic fibrosis, Angelman’s syndrome, and Liddle syndrome.
