Product Specific Information

Description:
The M1/70 monoclonal antibody reacts with mouse CD11b (integrin alphaM, 165–170 kDa). CD11b associates with CD18 to form the αMβ2 integrin (Mac-1), which binds CD54 (ICAM-1), C3bi, and fibrinogen.
Mac-1 is expressed on macrophages, NK cells, granulocytes, activated lymphocytes, and B-1 cells in the mouse peritoneal cavity. M1/70 also cross-reacts with human CD11b, allowing detection on human monocytes, granulocytes, and NK cell subsets.

Applications Reported:
Suitable for flow cytometric analysis.

Applications Tested:
Validated for flow cytometric analysis of mouse bone marrow cells. Use ≤0.25 µg/test (100 µL final volume). Optimize cell number empirically (10⁵–10⁸ cells/test).

Fluorochrome Information:
Brilliant Violet™ 480 (BV480) emits at 479 nm and is excited by a 405 nm violet laser. Ensure instrument compatibility.

Buffer Recommendation:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to minimize non-specific interactions.

Fixation Compatibility:
Not compatible with methanol-based fixation.

Target Information

CD11b (integrin alpha-M, ITGAM) is a 165 kDa adhesion molecule that forms a heterodimer with CD18 (integrin beta-2), known as Mac-1 or CR3. CD11b is expressed on monocytes/macrophages, granulocytes, activated lymphocytes, NK cell subsets, dendritic cells, and microglia.
It mediates adhesion via binding to ICAM-1, ICAM-2, ICAM-4, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X. Expression varies with cell type and maturation stage and is a common microglial marker.
CD11b dysfunction is associated with systemic lupus erythematosus (SLE) and ITGAM-related susceptibility to autoimmune disease.
