Product Specific Information
The 3D12 monoclonal antibody reacts with human CCR7 (CD197), also known as EBI-1. CCR7 is a G-protein-coupled chemokine receptor that binds CCL19 and CCL21. It is expressed on T cells and dendritic cells (DCs), distinguishing naive, central, and effector memory T cell subsets. CCR7 plays a role in T cell migration and DC maturation.
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometry using normal human peripheral blood cells.
- Recommended usage: 5 µL (0.5 µg) per test (100 µL staining volume).
- Incubation time: ≥45 minutes at 2–8°C.
Super Bright 780 Characteristics:
- Excitation: 405 nm
- Emission: 780 nm
- Recommended filter: 780/60 bandpass
- Light sensitive; protect from light.
- Use Super Bright Complete Staining Buffer (SB-4401) to minimize non-specific polymer interactions when using multiple Super Bright dyes.
Compensation Notes:
Higher compensation values may occur when using UltraComp eBeads (01-2222-42) compared to stained cells. In such cases, set compensation using cells. Similar results observed with AbC Total Antibody Compensation beads (A10497).
Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:
Super Bright Polymer Dyes are sold under license from Becton, Dickinson and Company.
Target Information
CCR7 is a G protein-coupled receptor of the chemokine family, induced by Epstein-Barr virus (EBV). It mediates EBV effects on B lymphocytes and is expressed in blood, bone marrow, lymph nodes, and intestines. CCR7 regulates migration of memory T cells and dendritic cell maturation. Its ligands include CCL19 and CCL21. Other related receptors include CCR1–CCR5 and the Duffy antigen, important for immune cell chemotaxis and inflammatory response.
For Research Use Only. Not for use in diagnostic procedures. Not for resale without express authorization.
