Product Specific Information

This F38-2E2 monoclonal antibody reacts with human CD366 (TIM3 or HAVCR2). CD366 is expressed on activated CD4+ T cell subsets (Th1, Th17, Treg), CD8+ T cells, monocytes, dendritic cells, and mast cells. It exists as membrane-bound and soluble forms due to alternative splicing. Galectin-9 is its ligand, and CD366 negatively regulates CD4+ T cells, influencing Th1 and Th17 cytokine secretion. It is implicated in tolerance, autoimmune disease (e.g., multiple sclerosis), and HIV infection.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometric analysis of stimulated human peripheral blood cells at 5 µL (0.06 µg)/test.
A test corresponds to staining a cell sample (10⁵–10⁸ cells) in 100 µL volume.

Super Bright 780 is a tandem dye excited at 405 nm and emits at 780 nm. Recommended filter: 780/60 bandpass. Ensure instrument compatibility.

When using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Compensation Notes:
Higher compensation values may occur in the violet 450/50 channel when using UltraComp eBeads (Product # 01-2222-42) or AbC Total Antibody Compensation beads (Product # A10497). Compensation with cells is recommended.

Light Sensitivity: Protect vial and samples from light.
Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333). Clone-specific performance should be tested empirically.

Target Information

TIM3 (HAVCR2) is a 281 amino acid, Type-1 Th1-specific cell surface glycoprotein expressed on terminally differentiated CD4+Th1 and CD8+Tc1 cells. It includes an IgV-like domain, a mucin-like domain, and a conserved tyrosine phosphorylation motif. TIM3 participates in macrophage activation, autoimmune disease induction, and immune tolerance regulation. It is associated with experimental autoimmune encephalomyelitis (EAE) and Th1-dependent autoimmune responses.
