Product Specific Information

The APB5 monoclonal antibody reacts with mouse CD140b, the beta chain of the platelet-derived growth factor receptor (PDGFRβ). PDGFRβ is a receptor tyrosine kinase that dimerizes upon ligand binding and phosphorylates substrates. CD140b is expressed in embryonic tissues and mesenchymal-derived adult mouse cells, playing a significant role in fibrous atherosclerotic lesion formation.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on NIH/3T3 cells at ≤1.0 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimal titration recommended.

Super Bright 780 is a tandem dye excited by the violet laser (405 nm) and emits at 780 nm. Recommended filter: 780/60 bandpass. Ensure instrument compatibility.

When using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity: Protect from light to prevent photo-induced oxidation.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness or FRET efficiency.

Target Information

PDGFRB is a receptor tyrosine kinase involved in atherogenesis and cell transformation by the SIS oncogene. It acts as a mitogen for mesenchymal-origin cells. The receptor can form homodimers or heterodimers with PDGFRA depending on ligand binding. The gene resides on chromosome 5 near GM-CSF and M-CSFR genes, implicated in 5-q syndrome. A translocation between chromosomes 5 and 12 fusing PDGFRB and ETV6 leads to chronic myeloproliferative disorder with eosinophilia.
