Product Specific Information

This 1RNM44N monoclonal antibody recognizes mouse CD170 (Siglec F), a cell surface lectin of the Ig superfamily binding sialic acid-containing glycoconjugates. The cytoplasmic domain includes an ITIM motif initiating intracellular signaling upon ligand binding. Siglec F is mainly expressed on eosinophils and alveolar macrophages, with lower levels on immature myelomonocytic cells. Mouse Siglec F is a functional ortholog of human Siglec 8 but not expressed on mast cells.

This antibody recognizes a formaldehyde-fixed epitope and is validated for flow cytometric analysis. Tested on mouse thioglycolate-elicited peritoneal exudate cells at ≤0.5 µg/test (100 µL final volume). Optimal titration recommended.

Super Bright 780 is a tandem dye excited by the violet laser (405 nm) and emits at 780 nm. Use a 780/60 bandpass filter and ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Compensation values for Super Bright 780 may vary between beads and cells; compensation with cells is recommended. Protect dye and samples from light due to photo-induced oxidation sensitivity.

Samples can be fixed and stored in IC Fixation Buffer (Product # 00-8222) or Fix/Lyse Solution (Product # 00-5333) up to 3 days at 4°C in the dark with minimal brightness loss.

Target Information

SIGLEC5 is a putative adhesion molecule mediating sialic acid-dependent binding. It binds both α2,3- and α2,6-linked sialic acids. Expression is highest in peripheral blood leukocytes, spleen, bone marrow, and lower in lymph node, lung, appendix, placenta, pancreas, and thymus. Found in monocytes and neutrophils, absent in early leukemic myelomonocytic cell lines.
