This antibody recognizes a monomorphic extracellular determinant of TCR alpha/beta, the dominant subtype of T cell receptor expressed in human peripheral blood.
Target Information
The T cell antigen receptor (TCR) consists of a ligand-specific alpha/beta heterodimer associated with invariant CD3 subunits (gamma, delta, eta, and zeta). These are essential for surface expression and signal transduction. Activation through the TCR leads to T cell differentiation, proliferation, and cytokine production.
Most T cells in blood and lymphoid organs express TCR alpha/beta heterodimers, while TCR gamma/delta T cells are mainly localized in epithelial tissues and infection sites.
Proper assembly of TCR-CD3 complex in the endoplasmic reticulum is required for surface expression; lack of any subunit prevents expression.
TCRs discriminate self from foreign peptides presented by MHC class II molecules, a process crucial for adaptive immunity. Dysregulation or recognition of self-peptides is linked to autoimmune diseases and tumors.
Superantigen engagement of TCRs can cause toxic shock syndrome. Autoantibodies against TCR V beta segments have been identified in autoimmune diseases such as rheumatoid arthritis and systemic lupus erythematosus, potentially modulating inflammatory responses.
Most human T cells express TCR alpha-beta with CD4 or CD8 (single positive), while a smaller subset lacks both markers (double negative). Increased alpha-beta double-negative T cells are observed in certain autoimmune and immunodeficiency disorders. Gamma-delta T cells, found mainly in epithelial tissues, exhibit limited diversity and distinct antigen recognition, with subsets showing antitumor and immunoregulatory activity.
