- Application Note: For IHC, epitope retrieval with citrate buffer pH 6.0 is recommended for FFPE tissue sections.
Target Information
The human AF9 gene is one of the most common fusion partner genes with the ALL1 gene at 11q23 (also called MLL), resulting in the translocation t(9;11)(p22;q23).
AF9 spans over 100 kb and contains two breakpoint cluster regions (BCRs):
- BCR1 within intron 4 (site A)
- BCR2 spanning introns 7 and 8 (site B)
Several structural elements have been identified in AF9, including:
- A colocalizing in vivo DNA topo II cleavage site
- An in vitro DNase I hypersensitive site in intron 7 (within BCR2)
- Two scaffold associated regions (SARs): SAR1 (intron 4) and SAR2 (exons 5–7)
These structural elements are shared with the MLL BCR. A DNA breakage and repair model for nonhomologous recombination between MLL and AF9 has been proposed.
