Target Information

The human AF9 gene is one of the most common fusion partner genes with the ALL1 (MLL) gene at 11q23, resulting in the t(9;11)(p22;q23) translocation.
AF9 spans more than 100 kb and includes two patient breakpoint cluster regions (BCRs):

• BCR1: within intron 4 (site A)
• BCR2: spans introns 7 and 8 (site B)

Structural elements identified in AF9:

• DNA topo II cleavage site (in vivo)
• DNase I hypersensitive site (in vitro, intron 7 in BCR2)
• Two scaffold-associated regions (SARs):
  • SAR1: intron 4
  • SAR2: exons 5–7

These regions correspond with breakpoint regions in leukemic cell lines.
A DNA breakage and repair model for nonhomologous recombination between MLL and AF9 has been proposed.
