Target Information
The human AF9 gene is one of the most common fusion partner genes with the ALL1 (MLL) gene at 11q23, resulting in the translocation t(9;11)(p22;q23).
AF9 spans more than 100 kb and includes two patient breakpoint cluster regions (BCRs):
- BCR1: within intron 4 (site A)
- BCR2: spans introns 7 and 8 (site B)
Several structural elements have been identified in AF9:
- DNA topo II cleavage site and DNase I hypersensitive site in intron 7 (BCR2)
- Two scaffold-associated regions (SARs): SAR1 (intron 4) and SAR2 (exons 5–7)
These structural features are shared with the MLL breakpoint cluster region, supporting a DNA breakage and repair model for nonhomologous recombination between MLL and AF9.
